Leptin Enhances Nitric Oxide Production and Decreases Blood Flow in Rat Skeletal Muscle

Objective: There are a few studies related to nitric oxide (NO) and leptin interaction in the regulation of physiological events skeletal muscle. Therefore, aim present study is investigate NO response blood flow synthase (NOS) distribution on rat Materials Methods: Twenty-four adult-male Wistar albino rats were divided into 4 groups: control (C), Leptin (LP) (50 μg/kg), L-NG-nitroarginine methyl ester (LN) (a non-specific inhibitor, 10 mg/kg,) L-NAME+Leptin (LN+LP) administrated groups. Drugs administered via right jugular vein. Hemodynamic parameters: mean arterial pressure, heart rate recorded during experiment at end samples for biochemical analyses nitrite/nitrate levels gastrocnemius muscle tissues from hindlimb NOS taken. Results: infusion after L-NAME administration significantly decreased (p<0.05, p<0.001). In addition, there was no change pressure group leptin-administered group. highest LP+LN (p<0.01). Decreased nitrate/nitrate with (p<0.01 vs C) reached values by infusion. Both endothelial (eNOS) neuronal (nNOS) distributions observed cells Conclusion: Although synthesis inhibited L-NAME, it concluded that partially enhances production uses as mediator its effects.
